Effect of Curcumin on Glucose Transport in Conditionally Immortalized Rat Brain Capillary Endothelial Cell line (TR-BBB) In Vitro

Abstract
Glucose is the important role of energy production as metabolic fuel in brain. The lack of glucose in brain gives rise to disease like hypoglycemia and brain coma etc. Curcumin, an active component of Indian spice turmeric, is known to have antioxidant and anti-inflammatory properties, preventing of Alzheimer¡¯s disease (AD) and induces apoptosis in many cancer cells. Also, it is reported that curcumin lowers blood glucose levels in diabetic mice and rats. In the present study, we studied the effect of curcumin on glucose transport in conditionally immortalized rat brain capillary endothelial cell line (TR-BBB) in vitro. We measured uptake of [©øH]3-O-methyl-D-glucose ([©øH]3-OMG) in TR-BBB to investigate changes of glucose transport across BBB by curcumin. As a result, [©øH]3-OMG uptake activity which was concentration-dependent with a Michaelis-Menten contstant of 20.6 mM and maximum rate of uptake (V_(max)) of 62.0 nmol/ (min ¡¤ mg protein) in TR-BBB. The uptake of [©øH]-3-OMG in TR-BBB was induced as increasing the concentration of curcumin pretreatment. In conclusion, curcumin is able to increase glucose uptake through the blood-brain barrier (BBB) and change level of glucose concentration in brain.
